HER3 (Phospho-Tyr1222)Colorimetric Cell-Based ELISA Kit

A cell-based ELISA from Assay Genie designed to measure phosphorylated HER3 protein at tyrosine 1222 in cultured human cells. The indirect ELISA format uses phospho-specific primary antibodies with HRP-conjugated secondary antibodies that bind the Fc region of the primary, catalyzing a colorimetric reaction upon substrate addition. Detection is read at 450 nm, with a dynamic range of > 5000 Cells.

The assay includes an anti-GAPDH monoclonal antibody that serves as an internal positive control for normalizing target absorbance values, plus a Crystal Violet whole-cell staining step for determining cell density. It is intended for measuring relative levels of HER3 phosphorylation in cultured cells and for screening the effects of treatments, inhibitors such as siRNA or chemicals, or activators on HER3 phosphorylation, and is suitable for use with adherent cells.

HER3, gene symbol ERBB3, is a receptor tyrosine kinase of the EGFR family that binds and is activated by neuregulins and NTAK. Although its kinase domain lacks catalytic activity, it heterodimerizes with other EGFR family members to transduce growth signals, and elevated expression has been reported in breast and other tumors, where it is indicative of poor outcome.
