HER3 Colorimetric Cell-Based ELISA Kit

A colorimetric cell-based ELISA kit from Assay Genie enables detection of HER3 directly in cultured cells without lysate preparation, providing a high-throughput workflow suited to adherent cell samples. The assay uses an indirect ELISA format: HER3-specific primary antibodies capture the target within fixed cells, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyse a colorimetric reaction upon substrate addition, read at 450 nm.

HER3, also known as ErbB3, is a member of the EGFR family of receptor tyrosine kinases involved in cell growth and survival pathways. The kit is reactive with human, mouse and rat samples, with a dynamic range of greater than 5000 cells per well. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method may be used to determine cell density, allowing the HER3 signal to be normalised to cell amount and plating differences to be adjusted.

The kit is provided in a 96-well microplate format and is stored at 4°C with a stability of 6 months. It can be used to measure relative HER3 levels in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), and activators on HER3 expression.
