HER2 (Phospho-Tyr1112)Colorimetric Cell-Based ELISA Kit

A cell-based ELISA from Assay Genie is designed to detect phosphorylated HER2 at Tyr1112 in cultured cells. The kit uses an indirect ELISA format in which HER2 (Phospho-Tyr1112) is captured by phospho-specific primary antibodies and detected with HRP-conjugated secondary antibodies that catalyze a colorimetric reaction at 450 nm upon substrate addition. The assay is described as convenient, lysate-free, high throughput and sensitive, and can be applied to measuring the relative amounts of phosphorylated HER2 in cultured cells as well as screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on HER2 phosphorylation.

Reactivity is reported for Human, Mouse and Rat samples, with adherent cells indicated as the sample type. The dynamic range is reported as > 5000 Cells. Because the Cell-Based ELISA is qualitative, two normalization methods are provided: a monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step can be used after the colorimetric read to determine cell density, allowing absorbance values to be normalized to cell amounts.

The target corresponds to ERBB2 (Gene ID 2064, UniProt P04626), a proto-oncogenic receptor tyrosine kinase of the EGFR family, and the kit is provided under the Phospho sub-type.
