Description

Assay Genie offers a cell-based ELISA designed for the detection of HDAC9 levels in cell lysates and culture supernatants, supporting research in the area of epigenetics and nuclear signaling with human samples. The kit uses a colorimetric detection method read at 450 nm in a 96-well microplate format, with a stated dynamic range of more than 5000 cells and a storage stability of 4°C for up to 6 months.

The workflow is built on an indirect ELISA format: HDAC9-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalization, while a Crystal Violet whole-cell staining step can be used to determine cell density and correct for plating differences. The kit provides a qualitative read-out of relative HDAC9 expression in cultured cells and is suited to screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HDAC9 levels.

HDAC9 is described as a transcriptional regulator of the histone deacetylase family, subfamily 2, that deacetylates lysine residues on the N-terminal part of core histones H2A, H2B, H3 and H4, and plays a role in transcriptional regulation, cell cycle progression and developmental events. The supporting database information lists Gene ID 9734, UniProt ID Q9UKV0, and a chromosomal location of 7p21.1 for the human ortholog. The target has been implicated in diseases including cancer, neurodegenerative disorders and cardiovascular diseases.
