HDAC6 (Phospho-Ser22)Colorimetric Cell-Based ELISA Kit

From Assay Genie, this Cell-Based ELISA Kit enables detection and quantification of HDAC6 phosphorylated at serine 22 directly in cultured cells. The assay uses a lysate-free, colorimetric workflow with measurement at 450 nm, offering a convenient and high-throughput approach to profiling HDAC6 phosphorylation in adherent cell cultures. Reactivity has been validated for Human and Mouse samples.

The kit employs an indirect ELISA format in which phospho-HDAC6 (Ser22) is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyse a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density and correct for plating differences. This makes the assay well-suited to measuring relative levels of phosphorylated HDAC6 as well as screening the effects of treatments, siRNA, chemicals or activators on HDAC6 phosphorylation.

HDAC6 is a histone deacetylase that modulates cellular processes including cell migration, cytoskeleton dynamics and protein degradation, and phosphorylation at serine 22 can influence its activity, making it a relevant target in studies of cancer, neurodegenerative disorders and inflammatory diseases. The dynamic range supports use with > 5000 cells per well.
