Description

From Assay Genie, this colorimetric cell-based ELISA kit is designed for the quantitative detection of HDAC5 (Histone Deacetylase 5) in cell lysates and tissue lysates. HDAC5 is described as a class II histone deacetylase that acts as a transcriptional regulator, deacetylating lysine residues on the N-terminal part of the core histones H2A, H2B, H3 and H4, and plays an important role in transcriptional regulation, cell cycle progression and developmental events. Dysregulation of HDAC5 has been linked to a variety of diseases, including cancer, cardiovascular diseases, and neurodegenerative disorders, making it a target of interest for therapeutic research.

The assay uses an indirect ELISA format in which HDAC5 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction measured at 450 nm after substrate addition. It is a lysate-free protocol intended for adherent cells, with a stated dynamic range of > 5000 Cells. A monoclonal anti-GAPDH antibody is supplied as an internal positive control for normalising target absorbance, and Crystal Violet whole-cell staining can be used to determine cell density. The kit shows reactivity with Human and Mouse samples and is supplied in a 96-Well Microplate format. Recommended storage is 4°C with a shelf life of 6 Months.

The kit can be used to measure relative HDAC5 expression in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HDAC5. Database cross-references listed for the target include UniProt IDs Q9UQL6 and Q9UKV0.
