Description

A cell-based ELISA kit from Assay Genie for the quantitative measurement of HDAC2 protein expression in cultured cells. The assay uses an indirect ELISA format in which HDAC2 is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies; the conjugated HRP catalyses a colorimetric reaction at 450 nm upon substrate addition. Described by the manufacturer as a convenient, lysate-free, high-throughput and sensitive assay, it can be used to measure the relative amounts of HDAC2 in cultured cells and to screen for the effects of inhibitors (e.g., siRNA or chemicals) or activators on HDAC2 expression.

Reactive with human, mouse and rat samples, with a stated dynamic range of > 5000 Cells, the kit is supplied in a 96-well microplate format. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and correct for plating differences. Storage is at 4°C, with a stated stability of 6 Months.

HDAC2 (histone deacetylase 2) is a member of the histone deacetylase family, subfamily 1, functioning as a transcriptional regulator. It catalyses the deacetylation of lysine residues on the N-terminal tails of core histones (H2A, H2B, H3 and H4) and acts via the formation of large multiprotein complexes, with roles in epigenetic repression, transcriptional regulation, cell cycle progression and developmental events. The human gene maps to chromosomal position 6q21 (Gene ID: 3066; UniProt ID: Q92769).
