Description

From Assay Genie, this cell-based ELISA kit is designed for the accurate detection of HDAC10 levels in cell lysates, tissue homogenates, and other biological samples. HDAC10 (histone deacetylase 10) is a key enzyme involved in the regulation of gene expression and chromatin remodeling, and its dysregulation has been linked to various diseases including cancer, autoimmune disorders, and neurological conditions, making it a valuable biomarker for investigating disease mechanisms and potential therapeutic targets.

The assay uses a colorimetric detection method read at 450 nm and is supplied in a 96-well microplate format with reactivity against human, mouse, and rat samples. The dynamic range is greater than 5000 cells, and the kit is intended for storage at 4°C with a stability of 6 months. It is categorized within the autophagy research area.

Qualitative determination of HDAC10 concentration is achieved through an indirect ELISA format: HDAC10 is captured by HDAC10-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. Following the colorimetric measurement, the Crystal Violet whole-cell staining method may be used to determine cell density, allowing absorbance values to be normalized to cell amounts and plating differences to be adjusted.
