Description

A cell-based ELISA from Assay Genie is designed for the sensitive and accurate detection of phosphorylated HDAC1 at serine 421 in cell samples. The kit uses a colorimetric detection method measured at 450 nm and is reactive with human, mouse, and rat samples, with a dynamic range of > 5000 cells. It employs an indirect ELISA format in which target-specific primary antibodies capture HDAC1 (Phospho-Ser421), and HRP-conjugated secondary antibodies bind the primary antibody's Fc region, catalysing a colorimetric reaction upon substrate addition.

A monoclonal GAPDH antibody is included as an internal positive control for normalisation, and Crystal Violet whole-cell staining is used to determine cell density. The lysate-free, high-throughput workflow supports qualitative measurement of relative phosphorylated HDAC1 levels in cultured cells and is suitable for screening the effects of treatments, inhibitors, or activators on HDAC1 phosphorylation. HDAC1 is a key enzyme in epigenetic regulation, deacetylating lysine residues on the N-terminal tails of histones H2A, H2B, H3, and H4, and its phosphorylation at serine 421 has been linked to cancer and neurodegenerative disorders.
