Description

A phospho-specific cell-based ELISA from Assay Genie designed for accurate detection of HBP1 phosphorylation at serine 402 in cultured cells, including adherent cells. The kit is a lysate-free, high-throughput assay that measures relative levels of phosphorylated HBP1 and can be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on HBP1 phosphorylation. It is intended for research into signal transduction pathways, drug discovery, and biomarker development within the research area of epigenetics and nuclear signaling.

The assay is configured as a cell-based, indirect ELISA with colorimetric detection at 450 nm. Phosphorylated HBP1 (Phospho-Ser402) is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies catalyse a colorimetric reaction upon substrate addition. Reactivity is human and the dynamic range is > 5000 Cells. Because the ELISA is qualitative, two normalisation methods are provided: a monoclonal anti-human GAPDH antibody serves as an internal positive control, and Crystal Violet staining may be used to determine cell density so absorbance values can be normalised to cell number.

HBP1 (UniProt O60381; Gene ID 26959) is a transcriptional repressor that binds target gene promoter regions and plays a role in regulation of the cell cycle and of the Wnt pathway, binding preferentially to the sequence 5'-TTCATTCATTCA-3'. It is a DNA-binding transcription factor located at chromosomal position 7q22.3 with a molecular weight of 57,645 Da, and is involved in Wnt receptor signalling, regulation of transcription, and cell cycle arrest.
