Description
Assay Genie supplies this Cell-Based ELISA Kit as a convenient, lysate-free, high throughput and sensitive solution for detecting HAT protein expression profiles in cultured cells. Using a qualitative indirect ELISA format, HAT-specific primary antibodies are paired with HRP-conjugated secondary antibodies to produce a colorimetric signal at 450 nm, with a dynamic range of > 5000 Cells. The kit is reactive with human, mouse and rat samples and is suitable for adherent cells as well as serum, plasma, lysate and cell culture supernatants, supporting measurement of relative HAT amounts and screening of treatments, inhibitors or activators.
For data normalization, the workflow incorporates a monoclonal anti-human GAPDH antibody as an internal positive control. After the HRP colorimetric readout, Crystal Violet whole-cell staining can be applied to determine cell density, allowing absorbance values to be adjusted for plating differences. Reagents are stored at 4°C, with stability maintained for six months.
The HAT target corresponds to histone acetyltransferase 1, a type B histone acetyltransferase involved in nucleosome assembly during DNA replication and repair. The manufacturer's overview notes that this protein participates in physiological processes including angiogenesis and cell proliferation, with dysregulation linked in the literature to cancer, cardiovascular disease and neurodegenerative disorders.
