Guinea pig TNF alpha ELISA Kit

From Assay Genie, this Sandwich ELISA kit is designed for the quantitative detection of tumor necrosis factor alpha (TNF-alpha) in guinea pig research samples. TNF-alpha is a key cytokine involved in the regulation of immune responses, inflammation, and cell apoptosis, and its dysregulation is linked to autoimmune disorders, inflammatory conditions, and infectious diseases. The kit uses a double-antibody sandwich format with colorimetric detection to deliver measurement of TNF-alpha in serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples.

Performance specifications include a sensitivity of 9.375 pg/mL and a working range of 15.625–1000 pg/mL. The kit should be stored at 2–8°C and remains stable for 12 months under those conditions. Validated linearity across serial dilutions gives ranges of 89–101%, 95–105%, and 85–102% for serum at 1:2, 1:4, and 1:8 dilutions respectively; 82–97%, 84–99%, and 84–98% for EDTA plasma; and 88–100%, 83–94%, and 84–99% for heparin plasma (n = 5 per sample type). Recovery testing reports average values of 97% for serum, 94% for EDTA plasma, and 92% for heparin plasma, with corresponding recovery ranges of 89–103%, 91–97%, and 89–98%.

The assay workflow involves equilibrating reagents and TMB substrate to room temperature, primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody incubation at 37°C for 60 minutes, HRP-Streptavidin binding at 37°C for 30 minutes, TMB color development in the dark for 10–20 minutes, and stop reaction with absorbance measurement at 450 nm.
