Guinea pig TIMP-2/Tissue Inhibitors of Metalloproteinase 2 ELISA Kit

Manufactured by Assay Genie, this double-antibody Sandwich ELISA kit enables the accurate measurement of TIMP-2 (Tissue Inhibitors of Metalloproteinase 2) in guinea pig samples. It uses a colorimetric detection method and is validated for serum, plasma, cell culture supernatant, cell or tissue lysate and other liquid samples. The target protein, identified by UniProt accession Q9WUC6, is also known by the synonyms CSC 21K, Metalloproteinase inhibitor 2, TIMP 2, TIMP2 and TIMP-2.

According to the product specification, the assay has a sensitivity of 0.188 ng/mL and a measuring range of 0.313-20 ng/mL. Linearity was demonstrated for serum (n=5), EDTA plasma (n=5) and heparin plasma (n=5) at 1:2, 1:4 and 1:8 dilutions, with percentage recoveries of 88-99%, 92-96% and 88-105% for serum; 91-103%, 85-105% and 83-96% for EDTA plasma; and 88-99%, 80-98% and 82-93% for heparin plasma. Spike-and-recovery testing gave averages of 94% for serum (range 87-104%), 95% for EDTA plasma (range 85-105%) and 98% for heparin plasma (range 91-102%).

The procedure includes equilibrating reagents and TMB substrate, a primary incubation at 37°C for 90 minutes, biotin-labelled detection antibody binding at 37°C for 60 minutes, HRP-Streptavidin (SABC) binding at 37°C for 30 minutes, TMB colour development in the dark for 10-20 minutes, and absorbance measurement at 450 nm after adding stop solution. The kit should be stored at 2-8°C for 12 months.
