Description

Assay Genie supplies a colorimetric, double-antibody sandwich ELISA kit developed for the quantitative measurement of PIIINP (N-Terminal Procollagen Propeptide) in guinea pig samples. The target, also referenced by the synonyms P3NP and Procollagen III Amino Terminal Propeptide, is recognised as a marker of collagen synthesis and degradation, supporting work in fibrosis, wound healing and tissue remodeling research.

The assay accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples, and is reported to deliver a sensitivity of 37.5 pg/mL across a standard range of 62.5-4000 pg/mL. Validation data describe linearity of 80-105% across 1:2, 1:4 and 1:8 dilutions of serum, EDTA plasma and heparin plasma, alongside recovery ranging from 85-101% (averages of 92% in serum and heparin plasma, and 97% in EDTA plasma).

The workflow involves equilibrating reagents to room temperature, loading standards, samples and controls onto the pre-coated plate and incubating at 37°C for 90 minutes, then adding a biotin-labeled detection antibody (37°C, 60 minutes), HRP-Streptavidin (37°C, 30 minutes), TMB substrate (10-20 minutes in the dark) and a stop solution, with absorbance read at 450 nm. Serum is prepared by clotting and centrifuging blood at 1000 × g for 20 minutes, while plasma is collected with anticoagulant and centrifuged at 1000 × g for 15 minutes at 2-8°C. Kits should be stored at 2-8°C for 12 months.
