Guinea pig NOS3/eNOS/Nitric Oxide Synthase 3, Endothelial ELISA Kit

Designed by Assay Genie for quantitative detection of endothelial nitric oxide synthase (NOS3/eNOS) in guinea-pig samples, this sandwich-format ELISA uses a double-antibody configuration with colorimetric readout. The kit is intended for research applications where NOS3 levels are investigated as a biomarker for conditions such as hypertension, cardiovascular disorders and diabetes, given the enzyme's central role in nitric oxide signalling and vascular physiology.

The assay has a sensitivity of 18.75 pg/mL and a working range of 31.25–2000 pg/mL, and accepts serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Storage is at 2–8°C for 12 months. Linearity was validated in serum, EDTA plasma and heparin plasma at 1:2, 1:4 and 1:8 dilutions, with recovery averaging 98% (serum), 95% (EDTA plasma) and 92% (heparin plasma), and recovery ranges of 86–105%, 85–105% and 88–103% respectively.

The procedure comprises reagent equilibration, a 90-minute primary incubation at 37°C, a 60-minute biotin-labelled detection antibody step, a 30-minute HRP-streptavidin binding step, TMB colour development in the dark for 10–20 minutes, and absorbance measurement at 450 nm after stop solution addition. The kit is supplied with a pre-coated microplate, lyophilised standard, biotin-labelled antibody (100×), HRP-streptavidin conjugate (SABC, 100×), TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer (25×), plate sealers and a technical manual.
