Description

From Assay Genie, this Sandwich ELISA uses a double-antibody format and colorimetric detection (read at 450 nm) to quantify MCP-1/CCL2 in guinea pig serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. MCP-1, also known as CCL2, is described as a chemokine involved in inflammation and immune cell recruitment, and is used as a biomarker in studies of inflammatory and autoimmune conditions.

Reported performance characteristics include a sensitivity of 9.375 pg/mL and a measuring range of 15.625-1000 pg/mL. Linearity has been evaluated at 1:2, 1:4 and 1:8 dilutions for serum (n = 5), EDTA plasma (n = 5) and heparin plasma (n = 5), with reported ranges of 95-104%, 90-99% and 90-99% for serum; 92-105%, 89-101% and 92-100% for EDTA plasma; and 90-105%, 91-98% and 90-99% for heparin plasma. Recovery ranges and averages across the same three sample types are 95-99% (97%), 95-101% (98%) and 95-105% (100%), respectively.

Unopened kits should be stored at 2-8°C for 12 months. The workflow consists of equilibrating reagents and TMB substrate to room temperature, loading standards, samples and blanks onto the pre-coated plate and incubating at 37°C for 90 minutes; adding biotin-labelled detection antibody with a 37°C, 60-minute incubation; adding HRP-Streptavidin (SABC) for 30 minutes at 37°C; developing with TMB substrate in the dark for 10-20 minutes; and adding stop solution before reading absorbance at 450 nm.
