Description

From Assay Genie, this sandwich ELISA kit is designed for the quantitative measurement of Brain-Derived Neurotrophic Factor (BDNF) in guinea pig samples. The double-antibody assay uses a colorimetric detection method with a reported sensitivity of 18.75 pg/mL and a working range of 31.25-2000 pg/mL. BDNF is described as a key neurotrophic factor involved in neuronal survival, growth, and differentiation, with dysregulation linked to neurological and mental health conditions.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Reported linearity in serum (n=5) is 86-101% at 1:2, 89-100% at 1:4, and 82-91% at 1:8; EDTA plasma (n=5) shows 91-103%, 84-100%, and 85-99%; and heparin plasma (n=5) shows 86-104%, 82-98%, and 83-99% across the same dilutions. Recovery ranges are 86-105% for serum (average 95%), 88-104% for EDTA plasma (average 95%), and 89-102% for heparin plasma (average 94%). The kit should be stored at 2-8°C for 12 months.

The procedure involves equilibrating reagents, a 90-minute primary incubation at 37°C, a 60-minute detection antibody incubation, a 30-minute HRP-streptavidin binding step, TMB color development in the dark for 10-20 minutes, and absorbance reading at 450 nm. Kit components include a dismountable ELISA microplate, lyophilized standard, biotin-labeled antibody concentrate, HRP-streptavidin conjugate, TMB substrate, sample and antibody dilution buffers, SABC dilution buffer, stop solution, wash buffer, plate sealer, and a technical manual.
