Description

From Assay Genie, this sandwich ELISA enables accurate colorimetric detection of Angiotensin-Converting Enzyme (ACE) 1 in guinea pig samples. ACE is a key enzyme in the renin-angiotensin system, converting angiotensin I to angiotensin II and playing a central role in blood pressure regulation and fluid balance. The kit supports research into cardiovascular disease, hypertension and related renal conditions, where ACE serves as a valuable biomarker for studying disease mechanisms and potential therapies.

Validated sample types include serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. Sensitivity is 0.188 ng/mL and the working range is 0.313–20 ng/mL. Reagents are stored at 2-8°C for 12 months. Reported linearity for serum (n=5) is 90-105% at 1:2, 89-99% at 1:4 and 85-103% at 1:8; EDTA plasma (n=5) ranges are 84-100%, 85-94% and 82-101%; heparin plasma (n=5) ranges are 82-96%, 82-96% and 80-95% across the same dilutions. Recovery is 89-105% for serum (average 97%), 85-105% for EDTA plasma (average 91%) and 85-105% for heparin plasma (average 92%).

The procedure equilibrates reagents and TMB substrate to room temperature, then incubates standards and samples at 37°C for 90 minutes. A biotin-labelled detection antibody is added for a 60-minute incubation at 37°C, followed by HRP-Streptavidin for 30 minutes at 37°C. TMB substrate is then incubated in the dark for 10–20 minutes, stop solution is added, and absorbance is measured at 450 nm.
