Guanylate Cyclase beta Colorimetric Cell-Based ELISA Kit

Assay Genie's cell-based ELISA kit is designed to measure guanylate cyclase beta protein expression in cultured cells. Using an indirect ELISA format with HRP-conjugated secondary antibodies and a colorimetric read-out at 450 nm, the kit allows qualitative determination of guanylate cyclase beta without the need for lysate preparation. It supports high-throughput screening of the effects that treatments, inhibitors (such as siRNA or chemicals) or activators have on guanylate cyclase beta expression.

The assay reacts with human, mouse and rat samples and is suitable for adherent cells. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalizing target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density. The dynamic range supports measurement of more than 5000 cells, with a recommended seeding of 20,000 adherent cells per well in a 96-well microplate format. The kit is stored at 4°C with a stability of 6 months.

Guanylate cyclase beta is the beta subunit of soluble guanylyl cyclase (gene symbol GUCY1B3), which catalyses the conversion of GTP to cyclic GMP, a signalling molecule involved in smooth muscle relaxation, neurotransmission, and cell growth. Dysregulation of the enzyme has been implicated in hypertension, heart failure, and erectile dysfunction, making it a target of interest in disease research and drug development.
