Description

This cell-based ELISA kit from Assay Genie is designed to detect GSK3beta (Phospho-Ser9) in cell lysates using an indirect ELISA format. The assay shows reactivity against human, mouse, and rat samples and uses adherent cells. Detection is colorimetric, measured at 450 nm, with a dynamic range of > 5000 cells.

The assay works by capturing the phosphorylated target with a phospho-specific primary antibody, followed by binding of an HRP-conjugated secondary antibody that catalyses a colorimetric reaction upon substrate addition. It is intended for studying relative levels of phosphorylated GSK3 in cultured cells and for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on GSK3 phosphorylation. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalize target absorbance values, and Crystal Violet whole-cell staining may be used to determine cell density for further normalization.

GSK3beta is a proline-directed protein kinase of the GSK family that phosphorylates and inactivates glycogen synthase, participates in the Wnt signaling pathway, and is involved in energy metabolism, neuronal cell development, and body pattern formation. Phosphorylation at Ser9 regulates its activity, making it a key signaling molecule in pathways related to cancer, diabetes, and neurodegenerative diseases.
