Description

From Assay Genie, this cell-based ELISA kit is designed for colorimetric detection of GSK3beta protein expression profiles in cultured cells using a convenient, lysate-free, high-throughput workflow. The assay is suited to adherent cells and is reactive with Human, Mouse, and Rat samples. Detection follows an indirect ELISA format, in which GSK3beta is captured by target-specific primary antibodies and visualized via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values. After the colorimetric measurement, a Crystal Violet whole-cell staining step can be used to determine cell density, allowing absorbance values to be normalized to cell amounts. The dynamic range is > 5000 Cells, with detection at 450 nm, and storage at 4°C with 6 months stability.

GSK3beta (glycogen synthase kinase-3 beta; Gene Symbol: GSK3B; Gene ID: 2932; UniProt ID: P49841; OMIM: 605004; Unigene: Hs.445733) is a proline-directed serine/threonine protein kinase of the GSK family with a molecular weight of 48,034 Da. It phosphorylates and inactivates glycogen synthase, participates in the Wnt signaling pathway, and is involved in energy metabolism, neuronal cell development, and body pattern formation. The kit supports research into GSK3beta's role in health and disease, with relevance to conditions such as Alzheimer's, diabetes, and cancer, and is suited to studies of signaling pathways, drug discovery, and disease mechanisms.
