Description

Designed for the accurate detection of phosphorylated GSK3α/β at Tyr279/216 in cells, this Cell-Based ELISA Kit from Assay Genie offers a high-throughput, lysate-free workflow with a colorimetric readout at 450 nm. The assay is reactive with human, mouse, and rat samples and can be applied to adherent cells, with a dynamic range of > 5000 cells.

The kit enables qualitative measurement of GSK3α/β phosphorylation and expression in cultured cells and can be used to screen the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on GSK3α/β phosphorylation. Detection is achieved through an indirect ELISA format, in which target-specific primary antibodies are bound by HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining step can be used to account for cell density.

GSK3α/β is a proline-directed kinase involved in various cellular processes, including cell growth, differentiation, and apoptosis, and phosphorylation at Tyr279/216 is an important regulatory event in multiple signalling pathways. Dysregulation of GSK3α/β has been linked to cancer, diabetes, and neurodegenerative disorders, and it is an active target for research and drug development, with known inhibitors including lithium and AR-A014418.
