Description

An In-Cell ELISA Kit from Assay Genie designed for the detection of G protein-coupled receptor kinase 7 (GRK7) levels in cultured cells. The assay uses a colorimetric detection method at 450 nm, is reactive with human samples, and has a dynamic range of > 5000 Cells. It is supplied in a 96-Well Microplate format and is intended to be stored at 4°C for 6 Months.

GRK7 is described as a key regulator of G protein-coupled receptor signaling pathways, and aberrant GRK7 activity has been linked to various diseases, including cardiovascular and neurological conditions, making it a relevant target for drug development. The kit provides a lysate-free, high-throughput workflow that detects the GRK7 protein expression profile directly in cells, and can be used both to measure the relative amounts of GRK7 in cultured cells and to screen the effects that various treatments, inhibitors (such as siRNA or chemicals), or activators have on GRK7.

Detection is achieved by an indirect ELISA format: GRK7 is captured by GRK7-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, with the conjugated HRP catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalising target absorbance values, and a Crystal Violet whole-cell staining method is used to determine cell density, allowing absorbance values to be normalised to cell numbers and plating differences to be adjusted. GRK7 itself is a retina-specific kinase involved in the shutoff of the photoresponse and in adaptation to changing light conditions via cone opsin phosphorylation, including rhodopsin.
