Description

From Assay Genie, this Cell-Based ELISA kit is designed for the analysis of GRK2 (G Protein-Coupled Receptor Kinase 2) phosphorylation at serine 685 in cell samples. It is a lysate-free, high-throughput and sensitive assay that uses an indirect ELISA format to detect GRK2 protein phosphorylation and expression profile in cells, making it suitable for measuring relative amounts of phosphorylated GRK2 in cultured cells as well as screening the effects that various treatments, inhibitors (such as siRNA or chemicals) or activators have on GRK2 phosphorylation.

The assay uses a colorimetric detection method at 450 nm, with HRP-conjugated secondary antibodies binding the Fc region of GRK2 (Phospho-Ser685)-specific primary antibodies to catalyze a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density for further normalization. The kit is reactive with Human, Mouse and Rat samples, with a dynamic range of > 5000 Cells, and is supplied in 96-well microplate format. It is intended for use with adherent cells.

GRK2 is a ubiquitous protein kinase of the GRK family that phosphorylates the beta-2-adrenergic receptor and related G-protein-coupled receptors, with expression level consistently elevated in chronic human heart failure. Phosphorylation of GRK2 at serine 685 has been implicated in cardiovascular disorders, diabetes and neurological conditions, making it a valuable target for drug development and disease research. The associated gene is ADRBK1 (Gene ID: 156, UniProt ID: P25098).
