Description

From Assay Genie, this Cell-Based ELISA Kit is designed for the accurate detection of phosphorylated GRK1 (G protein-coupled receptor kinase 1) at Ser21 in cell lysates, offering high sensitivity and specificity. GRK1 is a critical enzyme involved in the regulation of G protein-coupled receptor signaling, and phosphorylation at Ser21 is especially important for its activity and function; dysregulation has been linked to cardiovascular disorders, neurological conditions, and cancer. The kit supports research in the Signal Transduction area.

Reactivity covers Human, Mouse and Rat, with adherent cells as the supported sample type. The Sub Type is Phospho. The assay is lysate-free, high-throughput and sensitive, measuring the relative amounts of phosphorylated GRK1 in cultured cells and screening for the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators on GRK1 phosphorylation. Detection is colorimetric at 450 nm, with a dynamic range of > 5000 Cells.

Qualitative determination of GRK1 (Phospho-Ser21) is achieved by an indirect ELISA format in which target-specific primary antibodies capture the analyte and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, while the Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences.
