Description

From Assay Genie, this cell-based ELISA kit provides a convenient, lysate-free workflow for the detection of G protein-coupled receptor kinase 1 (GRK1) in cell lysates. Detection is colorimetric, measured at 450 nm, and uses an indirect ELISA format in which GRK1 is captured by specific primary antibodies and bound by HRP-conjugated secondary antibodies. The kit includes a monoclonal anti-GAPDH antibody to serve as an internal positive control for normalizing target absorbance, along with a Crystal Violet whole-cell staining step for determining cell density so that results can be adjusted for plating differences.

Reactive with human, mouse, and rat samples and suitable for adherent cells, the assay supports a dynamic range of > 5000 Cells. It is supplied in a 96-Well Microplate format and is intended for storage at 4°C for up to 6 months. The kit enables measurement of the relative amounts of GRK1 in cultured cells and supports screening of the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on GRK1, without the need for lysate preparation.

GRK1 is a serine/threonine kinase of the GRK family that phosphorylates and inactivates photon-activated rhodopsin and is implicated in phototransduction, rhodopsin-mediated signaling, and the regulation of G protein-coupled receptor signaling. Loss-of-function mutations in GRK1 are associated with Oguchi disease 2, a form of stationary night blindness, and dysregulation of GRK1 has been linked to cardiovascular, neurodegenerative, and metabolic conditions. The kit is categorized for signal transduction research and is suited to studies of cellular signaling, disease pathogenesis, and drug development.
