Granzyme B Colorimetric Cell-Based ELISA Kit

From Assay Genie, this colorimetric cell-based ELISA kit is designed to detect Granzyme B levels in cell lysates and tissue homogenates. The assay uses a qualitative indirect ELISA format in which Granzyme B is captured by target-specific primary antibodies, with HRP-conjugated secondary antibodies binding the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition.

Reactive with human, mouse and rat samples, the kit is a lysate-free, high-throughput and sensitive method for measuring relative Granzyme B protein expression in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals) or activators. Detection is read at 450 nm, with a dynamic range of more than 5000 cells.

A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, and a Crystal Violet whole-cell staining method may be used to determine cell density and adjust for plating differences. The kit is intended for adherent cells and should be stored at 4 °C, where it remains stable for 6 months.
