The GR (Glucocorticoid Receptor) Transcription Factor Activity Assay from Assay Genie is a DNA-binding immunoassay used to quantify activated GR binding to glucocorticoid response elements (GREs) in nuclear and cellular extracts. The microplate is pre-coated with GRE-containing double-stranded oligonucleotides, allowing only functionally active GR to be captured from nuclear lysates. Bound receptor is then detected with a GR-specific antibody and an HRP-based substrate, producing an absorbance signal proportional to DNA-binding activity in the sample.

This kit is applied in research on stress response, glucocorticoid signaling pathways, inflammation, immune regulation, and steroid hormone receptor pharmacology. It has been validated for human, mouse, and rat samples, supporting cross-species studies in cell line models and primary tissue. Typical workflows include preparation of nuclear extracts from treated cells, addition of 10-20 µL of lysate per well, and incubation steps of approximately 1-2 hours. The 96 Assays format provides sufficient wells for multiple treatment conditions, dose-response curves, and time-course experiments.

For best results, store all kit components at -20°C, protect from light, and avoid repeated freeze-thaw cycles of the antibody and oligonucleotide solutions. The assay is intended for research use only and is not approved for diagnostic or therapeutic applications.
