Description

This Sandwich ELISA kit from Assay Genie is intended for the quantitative measurement of Serum Amyloid A (SAA) in goat-derived samples. The double-antibody sandwich format is read colorimetrically at 450 nm following TMB substrate development. Compatible matrices include serum, plasma, cell culture supernatant, cell lysis buffer, tissue lysis buffer, and other biological fluids. The manufacturer reports a sensitivity of 0.938 ng/mL and a measurable range of 1.563–100 ng/mL, with kit components stable at 2–8°C for 12 months.

The assay workflow begins with reagent equilibration and a 90-minute primary incubation at 37°C, followed by a 60-minute biotinylated detection antibody step and a 30-minute HRP-Streptavidin (SABC) incubation. TMB substrate is then added for 10–20 minutes in the dark before the reaction is stopped and absorbance is read at 450 nm.

For sample preparation, serum is obtained by allowing blood to clot and centrifuging at 1000 × g for 20 minutes; plasma is collected in anticoagulant tubes and centrifuged at 1000 × g for 15 minutes at 2–8°C; tissue homogenates are prepared in PBS with protease inhibitors; cell culture supernatant is centrifuged at 2500 rpm for 5 minutes; and cell lysates are processed with lysis buffer containing protease inhibitors. Linearity at 1:2, 1:4, and 1:8 dilutions (n=5) ranges from 83–99% in serum, 82–102% in EDTA plasma, and 80–104% in heparin plasma. Mean sample recoveries are 96% (serum), 93% (EDTA plasma), and 94% (heparin plasma).
