Description

From Assay Genie, this Sandwich ELISA kit uses a double antibody format for the quantitative detection of IL-1B (interleukin-1 beta) in goat samples, including serum, plasma, cell culture supernatant, cell or tissue lysate, and other liquid samples. The kit targets IL1B, a key cytokine involved in the inflammatory response and immune regulation with roles in inflammation, autoimmune diseases, and infectious diseases. Sensitivity is 9.375 pg/mL with a working range of 15.625-1000 pg/mL (Uniprot P79162).

The assay protocol runs through six steps: equilibrate reagents and TMB substrate to room temperature, set standards, samples and controls on the pre-coated plate; incubate at 37°C for 90 minutes for antigen binding; add biotin-labeled detection antibody and incubate at 37°C for 60 minutes; add HRP-Streptavidin (SABC) and incubate at 37°C for 30 minutes; develop with TMB substrate in the dark for 10-20 minutes; add stop solution and read absorbance at 450 nm immediately.

Sample preparation: serum is clotted then centrifuged at 1000 × g for 20 minutes; plasma is collected with anticoagulant and centrifuged at 1000 × g for 15 minutes at 2-8°C; tissue is homogenized in PBS with protease inhibitors; cell culture supernatant is centrifuged at 2500 rpm for 5 minutes; cells are lysed using lysis buffer with protease inhibitors. Linearity at 1:2, 1:4, 1:8 dilutions: serum 87-105%, 84-95%, 86-100%; EDTA plasma 85-104%, 83-100%, 81-100%; heparin plasma 85-103%, 85-100%, 82-95%. Recovery: serum 86-101% (average 91%), EDTA plasma 91-105% (average 97%), heparin plasma 89-105% (average 97%). Storage is at 2-8°C for 12 months.
