Description

Designed by Assay Genie as a Cell-Based ELISA, this colorimetric kit enables sensitive detection of phosphorylated glycogen synthase at serine 645 directly in cultured cells, without the need for lysate preparation. The assay uses an indirect ELISA format in which target-specific primary antibodies capture Glycogen Synthase (Phospho-Ser645) and HRP-conjugated secondary antibodies generate a colorimetric signal measurable at 450 nm.

The kit reacts with human, mouse, and rat samples and is suitable for adherent cells. It supports a dynamic range of > 5000 Cells, allowing high-throughput screening of treatments, inhibitors, or activators and their effects on glycogen synthase phosphorylation. A monoclonal anti-GAPDH antibody is included as an internal positive control, and Crystal Violet whole-cell staining can be used to normalize absorbance values to cell density.

Glycogen synthase is a key enzyme in glycogen biosynthesis, transferring glucosyl residues from UDP-glucose to glycogen, and its phosphorylation at serine 645 is regulated by glucose-6-phosphate and PKA-mediated phosphorylation. Dysregulation of this phosphorylation event has been implicated in metabolic disorders such as diabetes and obesity, and the corresponding gene GYS1 (UniProt P13807) is associated with Glycogen Storage Disease 0, Muscle. The kit therefore offers researchers a tool for investigating metabolic pathways and potential therapeutic targets within the metabolism research area.
