Description

Assay Genie's cell-based ELISA kit is intended for the qualitative detection of GLUT3 protein expression directly in cultured adherent cells. The kit uses an indirect ELISA format in which GLUT3 is captured by GLUT3-specific primary antibodies and detected with HRP-conjugated secondary antibodies, yielding a colorimetric readout at 450 nm. A monoclonal anti-GAPDH antibody serves as an internal positive control, and a Crystal Violet whole-cell staining step is used to determine cell density, allowing target absorbance values to be normalized to cell amount.

Designed for use with human samples, the assay has a dynamic range of greater than 5000 cells and is provided in a 96-well microplate format with storage at 4°C and stability of six months. It supports measurement of relative GLUT3 levels in cultured cells and screening of treatments, inhibitors (including siRNA or chemicals), or activators for their effects on GLUT3 expression, making it relevant to research in signal transduction, glucose transport and cellular energy metabolism, with applications in metabolic disorders such as diabetes and cancer where GLUT3 dysregulation is implicated.

GLUT3, encoded by the SLC2A3 gene (Gene ID: 6515; UniProt ID: P11169; OMIM: 138170), is an integral membrane facilitative glucose transporter of the SLC family with a molecular weight of 53924 Da. One of thirteen human members of the equilibrative glucose transport protein family, its expression in humans is restricted to brain, testis (in spermatozoa), and skeletal muscle. Its surface expression is induced by insulin, and it mediates D-glucose transmembrane transport and L-ascorbic acid metabolism. The gene is located at chromosomal position 12p13.3.
