Description

Assay Genie provides a phospho-specific Cell-Based ELISA designed to measure levels of phosphorylated GluR4 at Ser862 in adherent cell samples. GluR4 is a subunit of the AMPA-type glutamate receptor, a class of ionotropic glutamate receptors that serve as the predominant excitatory neurotransmitter receptors in the mammalian brain. Phosphorylation at Ser862 has been linked to receptor trafficking, synaptic plasticity, and neuroprotective effects, making this modification a key target for studying neurological processes and potential therapeutic interventions.

The kit uses a colorimetric detection method read at 450 nm in an indirect ELISA format, with HRP-conjugated secondary antibodies catalysing a colorimetric reaction upon substrate addition. It is validated for reactivity with Human, Mouse, and Rat samples and has a dynamic range of > 5000 Cells. Because the assay is qualitative, normalization is achieved through two approaches: a monoclonal antibody specific for human GAPDH, included as an internal positive control, and Crystal Violet whole-cell staining to determine cell density.

The encoded protein, glutamate receptor 4, is an integral membrane protein and ligand-gated ion channel (Gene ID: 2893, UniProt ID: P48058, Gene Symbol: GRIA4), with a molecular weight of 49,146 Da. The gene is located on chromosome 11q22 and is subject to RNA editing (AGA->GGA; R->G) and alternative splicing that generates flip and flop isoforms. This kit is intended for Signal Transduction research.
