Description

Manufactured by Assay Genie, this Cell-Based ELISA Kit enables sensitive, high-throughput detection of Glucokinase Regulator (GCKR) protein expression directly in cultured cells without the need for lysate preparation. The colorimetric assay uses an indirect ELISA format in which GCKR is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. It is intended for research into diabetes, metabolic disorders, and drug discovery, supporting both qualitative measurement of GCKR levels and screening of treatments, inhibitors (such as siRNA or chemicals), and activators.

Assay features include a monoclonal anti-GAPDH antibody supplied as an internal positive control for normalizing target absorbance values, alongside a Crystal Violet whole-cell staining method for determining cell density. Reactivity covers human, mouse, and rat samples, with a dynamic range of greater than 5,000 cells. Detection is read at 450 nm, and the kit is supplied in a 96-well microplate format with a recommended storage temperature of 4 °C and a stability of 6 months.

GCKR is a key regulator of glucokinase in the liver and pancreas, with a molecular weight of 16,477 Da. The protein localizes predominantly to the cytoplasm when expressed alone and translocates to the nucleus upon co-expression with glucokinase.
