Description

This colorimetric cell-based ELISA kit from Assay Genie is designed for the qualitative detection and relative quantification of glucagon in cultured cells. The assay is reactive with human, mouse, and rat samples and is suitable for adherent cells. It operates as a lysate-free, high-throughput format and can be used to measure the relative amounts of glucagon in cultured cells as well as to screen the effects of various treatments, inhibitors, or activators on glucagon expression.

The kit employs an indirect ELISA format in which glucagon-specific primary antibodies capture the target, and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction at 450 nm upon substrate addition. A monoclonal anti-GAPDH antibody is included to serve as an internal positive control for normalising target absorbance values. Following the colorimetric measurement, Crystal Violet whole-cell staining can be used to determine cell density, allowing absorbance values to be normalised to cell amounts and adjusting for plating differences. The dynamic range is > 5000 cells, the format is a 96-well microplate, and storage stability is 4°C for 6 months. Glucagon (gene symbol GCG, Gene ID 2641, UniProt ID P01275) is a pancreatic hormone that counteracts the glucose-lowering action of insulin by stimulating glycogenolysis and gluconeogenesis, supporting research into metabolic disorders such as diabetes and hypoglycemia.
