Description

Manufactured by Assay Genie, this cell-based ELISA kit is designed for the accurate measurement of GIRK1 (also known as KIR3.1 or KCNJ3) levels in cell lysates and culture supernatants. GIRK1 is a G-protein-controlled potassium ion channel protein that plays a key role in regulating cellular excitability and membrane potential, with dysregulation implicated in neurological disorders including epilepsy and Parkinson's disease. The assay is intended for adherent cell samples and reacts with human, mouse, and rat material.

The kit uses a lysate-free, high-throughput indirect ELISA format with a colorimetric detection method read at 450 nm. It can be used to quantify relative GIRK1 protein expression in cultured cells and to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators. The reported dynamic range is > 5000 cells, and reagents are stored at 4°C for 6 months. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalization, while a Crystal Violet whole-cell staining step can be used to determine cell density after the colorimetric HRP/substrate reaction.

Intended for signal transduction research, the kit relies on HRP-conjugated secondary antibodies that bind the Fc region of the GIRK1-specific primary antibody and catalyze a colorimetric reaction upon substrate addition. The corresponding gene is KCNJ3 (Gene ID: 3760; UniProt ID: P48549).
