General Melatonin (MT) ELISA Kit

The General Melatonin (MT) ELISA Kit from Assay Genie is a competitive immunoassay for quantitative determination of melatonin in research samples. Melatonin (N-acetyl-5-methoxytryptamine) is the principal hormone secreted by the pineal gland and plays a central role in circadian rhythm regulation, seasonal biological cycles, and sleep-wake homeostasis. Investigators studying chronobiology, neuroendocrinology, sleep disorders, jet lag, shift work fatigue, or reproductive seasonality rely on accurate plasma melatonin profiling across the 24-hour light-dark cycle.

The competitive ELISA configuration uses a pre-coated microplate. Standards and unknowns are co-incubated with anti-melatonin antibody and biotinylated melatonin conjugate, followed by streptavidin-HRP and TMB substrate development. The recommended sample volume is 50 uL per well. No significant cross-reactivity has been observed with serotonin, N-acetyl-serotonin, 5-methoxytryptamine, or 6-hydroxymelatonin at physiological concentrations, supporting reliable discrimination of endogenous melatonin. Unopened kit components should be refrigerated; unused strips should be resealed with desiccant to maintain stability through the indicated shelf life.

Reagents provided include wash buffer concentrate, assay diluent, detection antibody, HRP conjugate, TMB substrate, stop solution, a microplate pre-coated with goat anti-rabbit IgG, lyophilized melatonin standard, and biotinylated melatonin. Reconstituted standards and conjugate are not stable to repeated freeze-thaw cycles; aliquot before freezing if extended storage is required. The kit is intended for research use only and is not validated for diagnostic or clinical procedures.
