Description

A Cell-Based ELISA kit from Assay Genie, designed for the precise measurement of GATA6 levels in cell lysates and cell culture supernatants. It provides a convenient, lysate-free, high throughput and sensitive assay that can detect GATA6 protein expression profiles in cells, and can be used both for measuring relative amounts of GATA6 in cultured cells and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on GATA6. Qualitative determination of GATA6 concentration is achieved by an indirect ELISA format, in which GATA6 is captured by GATA6-specific primary antibodies while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, allowing the conjugated HRP enzyme to catalyse a colorimetric reaction upon substrate addition.

Reactivity covers human, mouse and rat samples, with a Sample Type of Adherent Cells. Detection is carried out by a colorimetric method read at 450 nm, with a dynamic range of > 5000 Cells. The kit is supplied in a 96-Well Microplate format and is intended to be stored at 4°C with a stated stability of 6 Months. A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control in normalising target absorbance values, and following the colorimetric measurement of HRP activity the Crystal Violet whole-cell staining method may be used to determine cell density, allowing results to be analysed by normalising absorbance values to cell amounts and adjusting for plating differences. The research area is Epigenetics and Nuclear Signaling.

GATA6 is a transcription factor that plays a critical role in cell differentiation and tissue development, and is involved in physiological processes including cardiac development, gastrointestinal development and stem cell maintenance. Dysregulation of GATA6 has been linked to a range of diseases, such as congenital heart defects and gastrointestinal disorders, making it an important target for research and therapeutic development.
