Description

A Cell-Based ELISA Kit from Assay Genie designed for the accurate detection of phosphorylated GATA4 at Serine 262 in cell lysates. The colourimetric assay offers a convenient, lysate-free, high-throughput and sensitive format for measuring relative levels of phosphorylated GATA4 in cultured cells, and for screening the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on GATA4 phosphorylation.

Detection is achieved by an indirect ELISA format in which GATA4 (Phospho-Ser262) is captured by target-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody and catalyse a colourimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control to normalise target absorbance values, and the Crystal Violet whole-cell staining method may be used after substrate addition to determine cell density and adjust for plating differences.

The kit has a dynamic range of > 5000 cells, a colourimetric detection method read at 450 nm, and reactivity with human, mouse, and rat samples. Phosphorylation of GATA4 at Ser262 is known to play a critical role in cardiac development and function, with dysregulation linked to various cardiac diseases, making this a valuable tool for researchers studying gene expression regulation and potential therapeutic interventions in cardiac biology.
