Description

From Assay Genie, this cell-based ELISA kit is designed for the detection of GATA1 protein phosphorylated at serine 310 in cultured cells. Phosphorylation at this site is described in the literature as influencing GATA1 activity, making the kit relevant to studies of cell signalling, hematopoiesis, gene regulation and cell differentiation. The assay uses an indirect ELISA format: phospho-GATA1 (Ser310) is captured by target-specific primary antibodies and detected using HRP-conjugated secondary antibodies, with a colorimetric substrate read at 450 nm.

Qualitative measurement of GATA1 phosphorylation is supported in a lysate-free, high-throughput workflow suitable for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on GATA1 phosphorylation. An anti-GAPDH antibody is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step can be used to determine cell density. Reactivity has been established for human, mouse and rat samples, with a dynamic range of > 5000 Cells.

GATA1 is a member of the GATA family of transcription factors (UniProt P15976; NCBI Gene ID 2623) and is described as a general switch factor for erythroid development, binding the DNA consensus sequence 5'-[AT]GATA[AG]-3' within regulatory regions of globin and other erythroid genes. The protein has a reported molecular weight of 34,232 Da. The kit falls within the Epigenetics and Nuclear Signaling research area.
