Description

From Assay Genie, this cell-based ELISA kit is designed for the accurate measurement of GAD1 (glutamate decarboxylase 1) levels in cell samples, providing a high throughput, lysate-free workflow suited to neuroscience research. GAD1 catalyses the production of the inhibitory neurotransmitter gamma-aminobutyric acid (GABA), and its dysregulation has been implicated in neurological conditions including epilepsy, schizophrenia, and stiff man syndrome.

The assay uses an indirect ELISA format in which GAD1 is captured by target-specific primary antibodies and detected with HRP-conjugated secondary antibodies. Substrate addition produces a colorimetric reaction read at 450 nm. Reactivity covers human, mouse, and rat samples, and the dynamic range is stated as > 5000 cells. The kit is supplied in a 96-well microplate format, with storage at 4°C and a stated shelf life of 6 months.

For data normalisation, a monoclonal anti-GAPDH antibody is included as an internal positive control, and Crystal Violet whole-cell staining is used to determine cell density. Qualitative determination of GAD1 concentration is achieved by reading absorbance and normalising to cell amount, allowing the kit to be used for screening the effects of treatments, inhibitors (such as siRNA or chemicals), or activators on GAD1.
