Description

From Assay Genie, this Cell-Based ELISA Kit enables the accurate detection of phospho-tyrosine levels at position 643 of the GAB2 protein in cell lysates and tissue homogenates. Using an indirect ELISA format with colorimetric detection at 450 nm, the assay captures phosphorylated Gab2 with phospho-specific primary antibodies, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition.

The kit is validated for Human, Mouse, and Rat reactivity and supports a dynamic range of > 5000 Cells. Designed as a lysate-free, high-throughput assay suitable for adherent cells, it can measure relative phosphorylated Gab2 levels in cultured cells and screen for the effects of various treatments, inhibitors, or activators on Gab2 phosphorylation. Normalization is supported by an included monoclonal GAPDH antibody as an internal positive control, alongside a Crystal Violet whole-cell staining method to determine cell density after the colorimetric measurement.

GAB2 (UniProt ID: Q9UQC2) is an adaptor protein of the GRB2-associated binding protein family that plays important roles in regulating the growth, differentiation, and function of many hematopoietic cell types. Phosphorylation of tyrosine 643 is known to regulate GAB2 function and its downstream signaling pathways, and dysregulation of GAB2 has been implicated in various diseases, including cancer and neurological disorders.
