Description

This cell-based ELISA kit from Assay Genie is designed for the qualitative measurement of GAB2 (GRB2-associated-binding protein 2) in cultured cells. It uses a 96-well microplate format with a colorimetric detection method at 450 nm and is described as lysate-free, high throughput, and sensitive, with a stated dynamic range of more than 5000 cells. The kit is reactive with human, mouse, and rat samples and is intended for adherent cell culture.

The assay follows an indirect ELISA format: GAB2 is captured by target-specific primary antibodies, and HRP-conjugated secondary antibodies bind the Fc region of those primaries, catalysing a colorimetric reaction upon substrate addition. A monoclonal antibody specific for human GAPDH is included as an internal positive control for normalising target absorbance values, and a Crystal Violet whole-cell staining step may be used to determine cell density and adjust for plating differences. The kit can also be used to screen the effects of various treatments, inhibitors (such as siRNA or chemicals), or activators on GAB2. Storage is at 4 °C with a stated stability of 6 months.

GAB2 is an adaptor protein of the GRB2-associated binding protein family that plays important roles in regulating the growth, differentiation, and function of many hematopoietic cell types, and its dysregulation has been implicated in cancer, Alzheimer's disease, and diabetes. The human ortholog maps to chromosomal region 11q14.1 (gene ID 9846; UniProt Q9UQC2), and the encoded protein has a reported molecular weight of 69,968 Da.
