Description

Assay Genie provides this Cell-Based ELISA Kit for the quantitative measurement of phosphorylated GAB1 (Grb2-associated binder 1) at tyrosine 627 in cell lysates or tissue homogenates. The kit supports investigation of GAB1 phosphorylation levels, providing insight into signaling pathways involved in cell growth, differentiation, migration, proliferation, and survival. Phosphorylation at Tyr627 is essential for GAB1 activation and downstream signaling, making this target relevant to studies of intracellular cascades mediated by activated receptor-type kinases.

Detection is colorimetric, read at 450 nm, in an indirect ELISA format. Primary antibodies specific for the target capture phosphorylated GAB1, while HRP-conjugated secondary antibodies bind the Fc region of the primary antibody to catalyze a colorimetric reaction upon substrate addition. The assay is configured as a Cell-Based ELISA suitable for adherent cells, with reactivity established for Human and Mouse. The stated dynamic range is > 5000 cells.

For data normalization, the kit includes a monoclonal antibody specific for human GAPDH as an internal positive control. A Crystal Violet whole-cell staining method is supplied to determine cell density, allowing absorbance values to be normalized to cell amounts and adjusting for plating differences. The target gene is GAB1 (Gene ID: 2549; UniProt ID: Q13480), encoding GRB2-associated-binding protein 1, a member of the IRS1-like multisubstrate docking protein family, with a reported molecular weight of 80,005 Da.
