Description

A colorimetric cell-based ELISA from Assay Genie enables qualitative detection of G3BP-1 phosphorylation at serine 232 in cultured cells. The assay is reactive with human and mouse samples, employs colorimetric detection at 450 nm, and is rated for a dynamic range of > 5000 Cells. It uses an indirect ELISA format in which G3BP-1 (Phospho-Ser232) is captured by phospho-specific primary antibodies, with HRP-conjugated secondary antibodies catalysing a colorimetric reaction upon substrate addition.

The workflow is lysate-free and high throughput, supporting measurement of relative phosphorylated G3BP-1 levels and screening of the effects of treatments, inhibitors, or activators on G3BP-1 phosphorylation. A monoclonal anti-GAPDH antibody is supplied as an internal positive control for normalising absorbance values, and Crystal Violet whole-cell staining can be used to determine cell density for further normalisation. The sample type is adherent cells.

G3BP-1 is an hnRNA-binding protein and endoribonuclease involved in stress granule assembly and the Ras signal transduction pathway, and phosphorylation at S232 is linked to cellular stress responses relevant to cancer and neurodegenerative disorder research. The target is encoded by G3BP1 (Gene ID: 10146, UniProt ID: Q13283, OMIM: 608431, Unigene: Hs.587054), mapped to chromosomal location 5q33.1, with a reported molecular weight of 466.
