FOXO4 (Phospho-Thr451)Colorimetric Cell-Based ELISA Kit

Assay Genie offers a cell-based ELISA kit designed for the precise detection of phosphorylated FOXO4 at threonine 451 in cell lysates. The kit measures the relative amounts of phosphorylated FOXO4 in cultured cells and supports screening of various treatments, inhibitors (such as siRNA or chemicals), or activators on FOXO4 phosphorylation, making it useful for research into cancer, aging, and metabolic disorders where this phosphorylation event has been implicated.

Detection uses an indirect ELISA format in which FOXO4 (Phospho-Thr451)-specific primary antibodies are captured and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction read at 450 nm. The dynamic range is > 5000 Cells, and the kit is reactive with human and mouse samples. A monoclonal anti-GAPDH antibody is included as an internal positive control for normalisation, and a Crystal Violet whole-cell staining method can be used to determine cell density so that absorbance values can be adjusted for plating differences.

FOXO4 is a forkhead box transcription factor that regulates cell metabolism, apoptosis, and stress response. The protein is encoded by the FOXO4 gene (Gene ID 4303, UniProt P98177) with a molecular weight of 47,941 Da, and its human ortholog is located at chromosomal position Xq13.1.
