FOXO1A (Phospho-Ser256)Fluorometric Cell-Based ELISA Kit

From Assay Genie, this fluorometric cell-based ELISA kit is designed to monitor phosphorylation of FOXO1A at serine 256 and to profile its expression directly in cultured cells. The lysate-free, high-throughput workflow uses an indirect ELISA format: target phosphorylation is captured by FOXO1A (Phospho-Ser256)-specific primary antibodies, while Dye 1- and Dye 2-conjugated secondary antibodies enable a dual-channel fluorometric readout. The kit is reactive with human, mouse and rat samples and supports a dynamic range of > 5000 Cells.

It is suited both to measuring the relative amounts of phosphorylated FOXO1A in cultured cells and to screening how various treatments, inhibitors (such as siRNA or chemicals) or activators affect FOXO1 phosphorylation. Two normalization strategies are provided: a monoclonal anti-GAPDH antibody serves as an internal positive control for normalizing target RFU values, and an antibody against the non-phosphorylated counterpart of FOXO1A (Phospho-Ser256) is included so the RFU value for phosphorylated FOXO1A can be normalized against total FOXO1A. Plates are read at Ex/Em 651/667 (Dye 1) and 495/521 (Dye 2). The kit is supplied with storage at 4 °C and a stated stability of 6 months.
