Description

Assay Genie's colorimetric cell-based ELISA kit is designed for the precise measurement of FOXL1 levels in cell cultures. The assay is lysate-free and high throughput, using an indirect ELISA format in which FOXL1 is captured by target-specific primary antibodies and detected via HRP-conjugated secondary antibodies that catalyze a colorimetric reaction upon substrate addition. Results are read at 450 nm on a standard microplate reader.

A monoclonal antibody specific for human GAPDH is included to serve as an internal positive control for normalizing target absorbance values, and a Crystal Violet whole-cell staining step is provided to determine cell density so that absorbance values can be normalized to cell amounts. The kit can be used to measure relative FOXL1 expression in cultured cells as well as for screening the effects of treatments, inhibitors (such as siRNA or chemicals) or activators on FOXL1.

The plate format is a 96-well microplate suitable for adherent cells, with a stated dynamic range of greater than 5000 cells. The kit is intended for human samples and is stored at 4 °C with a stated stability of 6 months. FOXL1 (Forkhead box protein L1; UniProt Q12952, Gene ID 2300) is a forkhead/winged-helix-box transcription factor required for proper proliferation and differentiation in the gastrointestinal epithelium, and is a downstream target of hedgehog signalling via GLI2 and GLI3. Dysregulation of FOXL1 has been linked to cancer and gastrointestinal disorders, supporting its relevance as a research biomarker.
