FOXK1 Transcription Factor Activity Assay

This Assay Genie kit measures the DNA-binding activity of FOXK1 (Forkhead box protein K1), a forkhead/winged-helix transcription factor implicated in autophagy regulation, myogenic differentiation, and downstream mTOR pathway signaling.

The plate is pre-coated with a double-stranded oligonucleotide bearing the FOXK1 consensus binding sequence; nuclear or whole-cell lysates are added, and bound FOXK1 is detected with a target-specific primary antibody and an HRP-conjugated secondary antibody using a TMB substrate.

A typical reaction requires a 10–20 μL sample per well and the complete protocol runs in approximately 3–4 hours. Results are reported as relative activity (fold-change versus control) rather than absolute concentration, which makes the kit appropriate for signaling pathway studies in muscle biology, autophagy, and cancer research.

Reagents should be stored refrigerated at 4°C and kept away from light until use; the kit is supplied for research use only across human, mouse, and rat samples.
