FOXK1 Colorimetric Cell-Based ELISA

A Cell-Based ELISA Kit from Assay Genie designed for the detection of Forkhead box K1 (FOXK1) protein in adherent cells. The assay uses an indirect ELISA format in which FOXK1 is captured by target-specific primary antibodies and HRP-conjugated secondary antibodies bind the Fc region of the primary antibody, catalysing a colorimetric reaction upon substrate addition. The kit is lysate-free and supports high-throughput measurement of relative FOXK1 expression in cultured cells, and can be used to screen the effects of various treatments, inhibitors such as siRNA or chemicals, or activators on FOXK1 levels.

Detection is colorimetric at 450 nm, with a stated dynamic range of > 5000 cells. The format is a 96-well microplate, with storage stability of 4°C for 6 months, and reactivity covers human and mouse samples. Two normalisation approaches are supported: a monoclonal antibody specific for human GAPDH serves as an internal positive control for normalising target absorbance values, and the Crystal Violet whole-cell staining method can be used to determine cell density so absorbance values can be normalised to cell amounts and plating differences adjusted.

FOXK1 is described as a transcription factor (DNA-binding) with roles in myogenic differentiation and adult skeletal muscle remodelling in response to physiological stimuli, and it localises to the nucleus. The research area indicated for the kit is developmental biology.
